Derived from consenting donor cord blood and fibroblasts, our striatal neurons have been specifically developed for use in mono-, co- and tri-culture models of neurodegenerative disease and for drug discovery.  

The vast majority of human striatal neurons are γ-amino butyric acid (GABAergic) projection medium spiny neurons (MSNs). These cells typically express dopamine and cAMP-regulated phosphoprotein (DARPP32). It is these cells that specifically degenerate in the early phase of Huntington’s disease (HD), causing progressive loss of the patient’s motor and cognitive functions.

Axol has developed a medium formulation that can efficiently drive the differentiation of iPSC-derived neural stem cells to the striatal neuronal lineage. The neurons derived using the medium kit are functionally active and express the key markers of human MSNs (DARPP32, CTIP2, CALBINDIN, and GABA) and form functional synapses in twenty days. With our striatal neurons, you can build robust mono-, co- and tri-culture models with isogenic astrocytes, microglia and cortical inhibitory interneurons to investigate neurological diseases (Alzheimer’s, Parkinson’s, and Huntington’s Disease; epilepsy) and for drug screening and discovery.

Applications

Our axoCellsTM striatal neuron progenitors are suitable for investigating Huntington’s disease and for screening candidate therapies for this disease.

Healthy control lines are also available and can be used with highcontent imaging, MEA, microfluidic and organ-on-chip (OOC) devices to measure structural and functional responses in neuronal models.

Protocols

Fully optimized cell culture reagents are available for the differentiation of progenitors into mature striatal neurons within 21 days. In addition, single media protocols, enabling co- and tri-culture of our striatal neurons with inhibitory interneurons, astrocytes and microglia provide convenient and fast culture solutions to better model human diseases in vitro.
