How to seed your cardiomyocytes on MEA
In this video, you will learn how to prepare the MEA plate before seeding iPSC-derived Cardiomyocytes. We will walk you through setting up the cloning rings, coating with fibronectin and finally seeding cardiomyocytes on to the MEA plate.

How to prepare the MEA plate
Step 1: Prepare required materials
Step 2: Place the CellSpotter-comfort into an MEA 24-well plate-comfort pre-coated with Fibronectin 40 μg/mL
Step 3: In a sterile dish stand the cloning rings upright
Step 4: Test for stability of cloning ring by tapping the sterile dish. If the cloning ring falls over turn it round the other way.
Step 5: Add 100 μL of Plating Medium to each cloning ring to wet them
Step 6: Gently tap the cloning rings to remove the medium
Step 7: Remove the fibronectin from the MEA plate
Step 8: Add the wetted cloning rings to the CellSpotter installed on the MEA plate
Step 9: Add cells into the cloning ring
Step 10: Add Plating Medium into the well surrounding the cloning ring, repeat for each well
Step 11: Incubate the MEA plate overnight at 37oC, 5% CO2
Step 12: Remove the cloning rings
Step 13: Remove the CellSpotter
