Custom Manufacturing of iPSC-Derived Microglia to Advance Neurodegeneration Disease Models

Custom Manufacturing of iPSC-Derived Microglia to Advance Neurodegeneration Disease Models

At Axol, we’ve spent over a decade working and improving our approach to iPSC differentiation, helping researchers accelerate drug discovery and advance human-relevant disease models.

We employ our expertise in custom manufacturing to produce iPSC-derived microglia for models of neurodegenerative diseases such as Alzheimer’s Disease (AD), Amyotrophic Lateral Sclerosis (ALS), and Parkinson’s Disease.

iPSC-Derived Microglia for Better Human Disease Models

Microglia are the primary immune cells of the central nervous system. They play key roles in brain development, neurogenesis, synaptic pruning, and maintaining the normal homeostatic environment. Human iPSC-derived microglia are made by differentiating iPSCs from reprogrammed donor blood samples. They therefore retain the phenotypic characteristics of the donor, producing a more human-relevant model system. This also opens up exciting applications of disease-derived microglia to test potential therapies in specific human disease models.

The characteristics of our axoCells™ Microglia  go beyond morphology. We’ve validated their functionality through a range of biologically relevant assays. These include the expression of key markers, phagocytosis, chemotaxis and cytokine release.

If you want iPSC-derived microglia from other cell lines, our custom manufacturing services can be tailored to meet the unique requirements of your project. We can manufacture from our lines, your lines, or third-party lines.

Scalable Manufacturing of Functional iPSC-Derived Microglia

At Axol, we offer a scalable approach to functional cell manufacturing. Our approach is built on collaboration, and we are obsessed with quality. We work closely with our clients to ensure that their needs are met with precision and care.

We see the deliverable not ‘just’ cells but the essential quality, functional, consistent inputs to your in vitro system. We can do small runs or very large runs; we can synchronize with your program of work to ensure availability of stock. With scale comes quality, reproducibility and availability. Using our robust differentiation protocols, we are not just delivering cells; we’re helping to build more accurate, more reliable disease models.

Whether you want to scale up in size or complexity, run large screening platforms, or just want to ensure consistency for the future, we’ve developed the manufacturing capabilities to support your bespoke experimental designs.

We offer:

  • Protocols for using microglia in monoculture, or incorporating microglia into co- and tri-culture with neurons
  • Functional Characterization
  • Microglia from disease lines (including TREM2 Alzheimer’s Disease)
  • Manufacture of large batches (up to 250 x 1 million vials as standard, and more if needed)
  • Custom QC services to ensure cell quality and supply.

New Collection of iPSC lines from Patients with Sporadic Alzheimer’s Disease

In partnership with StrataStem, Axol Bioscience have released a new collection of iPSC lines generated from primary materials donated from patients with sporadic Alzheimer’s Disease (sAD). The collection consists of 20 iPSC lines, including sAD and unaffected donor controls. These lines are available for use for custom manufacturing of cortical excitatory and inhibitory neurons and astrocytes, as well as microglia.

  • Gender mixed, age relevant to Late-Onset AD.

Investigating TREM2 with gene-edited iPSC lines

As part of our ongoing work building advanced in vitro AD models, we have explored the role of TREM2 with gene-edited iPSC lines. TREM2 (triggering receptor expressed on myeloid cells 2) is a gene expressed by microglia in the brain and TREM2 mutations have been linked to the development of neurodegenerative diseases, including AD.

TREM2 microglia can be manufactured through our made-to-order ‘custom differentiation’ services.

TREM2 microglia can be manufactured through our made-to-order ‘custom differentiation’ services. from the following lines:

  • iPSC line CENSOi073-A Female, 41 yrs at time of sampling. Homozygous TREM2, c.150G>T p.(Trp50Cys)
  • iPSC line CENSOi075-B Female, 69 yrs at time of sampling. Heterozygous TREM2, c.150G>T p.(Trp50Cys)
  • iPSC line CENSOi078-A Male, 68 yrs at time of sampling. Heterozygous TREM2, c.150G>T p.(Trp50Cys)
